Kit to asses sperm morphology

A. USAGE:

• SPERM QUICK MORPHO help to differentiation of spermatozoa morphology with excellent staining for sperm head, neck and tail.

B. CONTENTS:

• Fixative solution (FS).
• Stain (A) solution (SAS).
• Stain (B) solution (SBS).
• Product insert.

C. PRECAUTIONS:

• All patient samples should be considered potentially infectious and the user must wear protective gloves, eye protection, face mask and laboratory coats when performing the test and take off contaminated parts immediately.
• After contact with skin wash immediately with water and soap.
• The test should be discarded properly after use (biohazard).
• Do not use after expiration date, which appears on the package label.

D. WARNINGS:

• Care should be taken to avoid contact with skin or eyes, and to prevent inhalation or swallowing.
• Work under air removal environment.
• Do not release the products used into the environment.
• Fixative and Stain (A) highly flammable liquid and vapor.

E. STORAGE:

• The kit keeps dry at 2 – 8 °C. tightly closed when not in use.

F. PEPARATION:

• The kit reagent is ready to use.

G. PROCEDURES:

• Transfer solutions into jars or staining dishes.
• Prepare thin layer of semen sample on slides after removing seminal plasma and add suitable buffered media (e.g.: DPBS or EBSS) for clear staining, then wait for complete air drying.
• Dip slides for 5 seconds in Fixative solution (FS), then wait for complete drying.
• Dip slides in Stain (A) solution (SAS) for 5 seconds, Blot slide on towel to remove excess stain before transferring to next step.
• Dip slides in Stain (B) solution (SBS) for 5 seconds, then dip slides one time in tap water for remove excess stain, then wait for complete air drying.
• Examine under light microscope at 100X lens using immersion oil, Spermatozoa acrosome stained with pink to violet color, while tail stained with blue to dark blue.
